| Abstract | Unlike ion channel modulators that act on natural ion channel proteins, synthetic ion channels are small organic molecules that self-assemble to form ion channels in cell membranes irrespective of the presence of natural ion channels. By using HPTS assay, Ussing chamber, and patch-clamp single channel recording, we have discovered a range of small molecules with unique physical properties and excellent ion selectivity. In addition, on the basis of structural studied using STD-NMR, NOESY-NMR and CD methods, we have proposed models of self-assembled ion channels and further evaluated them using all-atom molecular dynamics simulations. Those synthetic ion channels may offer great opportunity to resolve unmet medical needs. |
