Objectives
1 To determine MIC of Bi(III) tropolone against Staphylococcus aureus, and monitor time-dependent Bi(III) cellular uptake in bacterial cells 2 To identify Bi(III)-binding proteins in bacterial cells upon treatment of bacterium with sub-lethal concentration of Bi(III) tropolone by our established continuous-flow gel electrophoresis coupled with inductively-coupled plasma mass spectrometry 3 To profile Bi(III)-regulated proteins by MS-based quantitative proteomic approach 4 To uncover the biological pathways perturbed by Bi(III) tropolone and to explore protein-protein interaction network of Bi(III)-binding and regulated proteins to unveil hub proteins by bioinformatics 5 To subsequently validate key protein targets and the key biological pathways perturbed by Bi(III) tropolone through either bioassays or purified proteins using various biophysical and molecular biology methods
