Objectives
1 To further characterize the phenotypic changes of PDGFRα-Piezo1 KO mice, and to consolidate that Piezo1 deletion specifically in PDGFRα+ MSCs leads to reduced bone mass and increased marrow adiposity. 2 To interrogate whether Piezo1 in MSCs mediates exercise-induced bone formation, reduction in marrow adipocytes and improvement in systemic insulin sensitivity. 3 To investigate detailed intracellular signaling pathway by which Piezo1 controls the cell fate determination of MSCs. 4 To identify the Piezo1-dependent secretory factors that directs the differentiation of MSCs and function of bone marrow adipocytes.
