Objectives
1) To generate isogenic infectious clones of a MERS-CoV (Korean lineage) virus which have ether AA nsp6 232F or 232L; and to phenotype these isogenic viruses in regard to replication competence in 2) human respiratory epithelial cell lines, 3) ex vivo cultures of the human lung and bronchus and in 4) lungs of human DPP4 transgenic mice. The biological role of nsp6 L232F on double membrane vesicle formation and innate immune evasion will be investigated.
