Objectives
1 To comprehensively interrogate whether reduced β-klotho expression in pancreatic islets is a causal contributor to defective insulin secretion, β-cell loss, and hyperglycemia in type-2 diabetic mice; 2 To investigate whether β-klotho controls insulin secretion through modulation of glycolysis in pancreatic β-cells of type-2 diabetic mice; 3 To investigate whether the regulatory role of β-Klotho in glycolysis and GSIS is mediated by HIF1/ARNT or other transcription factors in pancreatic β-cells; 4 To elucidate the molecular and structural basis whereby β-Klotho induces HIF1a, glycolysis, and GSIS in pancreatic β-cells.
