Objectives
1 To identify bismuth binding proteins in both H. pylori and other pathogens by a home-made integrative approach consisting of a fluorescence probe and continuous flow gel electrophoresis ICP-MS (GE-ICP-MS); 2 To carry out quantitative proteomics to quantify those proteins up- and down-regulated by bismuth antimicrobial drugs in both H. pylori and related pathogns; 3 To carry out quantitative proteomics (iTRAQ) to quantify those proteins up- and down-regulated by bismuth antimicrobial drugs in both H. pylori and other pathogens (C. jejuni); 4 To identify important low molecular mass metabolites upon the drug treatment by NMR-based metabolomics and validate the key proteins/enzymes or biological pathways perturbed; 5 To verify the identified key targets identified by metalloproteomics and metabolomics by in vivo bioassay and/or in vitro characterization.
