Objectives
1 To perform molecular cloning of the putative protein targets of bismuth antimicrobial drugs, and to overexpress and purify the proteins including Ahpc, UreG, Fur and Dnak 2 To examine the in vitro binding properties including binding stoichiometry, affinity of bismuth drugs with the proteins by biochemical and biophysical techniques 3 To investigate the effect of bismuth drugs on the function of selected protein targets through bioassays 4 To further validate the potential disruption of bismuth drugs on the protein functions in vivo by gene knockout
