Objectives
食管鱗狀細胞癌(鱗癌)是世界各地包括香港在内最常見的食道惡性腫瘤。術前新輔助放化療已證實可以改善患者預後,然而,並不是所有患者都適合術前新輔助治療。除此之外,目前尚缺乏預測放化療療效的檢測方法。因此,本項目旨在研發一種血清學檢測方法用於食管癌術前放化療的療效預測。初步數據顯示食管癌耐药患者血清中存在高水平的microRNA-16 (miR-16) 和microRNA-193b (miR-193b)。據此,我們將深入探討血清miR-16和miR-193b水平作為放化療生物標誌物的可行性。首先,我們將建立拷貝數定量標準曲線用於計算miR-16和miR-193b的血清水平並進行大樣本驗證。其後,我們將通過檢測miR-16和miR-193b在臨床組織中的表達水平和在患者不同治療時期的血清中表達水平的變化,推測它們的來源。最後,利用食管癌順鉑耐药细胞模型研究miR-16和miR-193b水平與細胞耐藥性的關聯。本項目的順利完成,將有望開發出定量檢測血清miR-16和miR-193b水平的試劑盒,用於指導食管鱗癌患者的個體化治療。
