AbstractThe localization and transcriptional level of keratinocyte growth factor (KGF ) mRNA in normal (N), inflammatory (IF), dysplasia (DYS) and squamous cellcarcinoma (SCC ) were observed by in situ hybridization in order to determine the distribution and effects of KGF in normal and benign conditions and the carcinogenesis of laryngeal mucosa. The results showed that KGF mRNA was expressed by fibroblasts, endothelial cells and inflammatory cells, but not by any of the epithelial cells. There was an increasing tendency of mRNA transcriptional lever from N, IF, DYS to SCC. Furthermore, the expression level was higher in poorly differentiated SCC than in well differentiated SCC. It is concluded that KGF plays an important role in the interaction between epithelial cells and mesenchymal cells. KGF might be one of the critical factors for maintaining the structure and metabolism of llormal mucosaand the growth of laryngeal carcinoma. 利用原位雜交的方法檢測KGFmRNA在正常喉粘膜上皮(N)、慢性非特異性炎癥(IF)、不典型增生(DYS)及鱗癌(SCC)中的轉錄水平,探討KGF在喉粘膜良性及惡性病變中的分布和可能的作用。結果表明,KGFmRNA不僅在間質中的成纖維細胞中表達,少量的炎細胞及血管內皮細胞中亦表達,而且從N、IF、DYS到SCC、KGFmRNA轉錄水平逐漸增強;上皮細胞及腫瘤性上皮細胞不表達KGFmRNA,KGFmRNA在分化差的SCC周圍間質中表達較分化好的SCC周圍間質增多。結論:KGF在上皮與間充質細胞的交互作用中發揮著重要的作用,對維持喉粘膜正常結構、代謝及喉癌的發生發展具有重要意義。
