Objectives
To use the Lerman's MELT AND SQHTX programmes to simulate melting profile and electrophoretic behaviour of the DNA fragment(s) to be analysed. It will also assist in the design of the oligonucleotide primers and/or with the necessary GC clamps for DNA amplification in order to resolve all variations in each of the DNA fragments; to develop the denaturing gradient gel electrophoresis in Hong Kong for detecting variations in the G6PD gene; to use the DGGE to detect unequivocally female carrier/heterozygote and identify at risk families; to develop direct sequencing of some of the amplified products so as to identify new mutation(s).
