Objectives
1 Establish the DNA display technique on beads and investigate integration with emulsion PCR using previously discovered aptamers as a proof-of-principal. 2 Evolve new fluorophore-RNA aptamers with a range of selected fluorescent properties using fluorescence activated cell sorting coupled to deep sequencing. 3 Characterize newly selected fluorophore-RNA aptamers in vitro for biophysical properties, stability, environmental impact on fluorescence, and potential for fusion with longer RNAs. 4 Observe newly selected fluorophore-RNA aptamers expressed in cells using E. coli and human cell culture as two independent model cell systems.
