Objectives
3.1 To screen small molecular compounds that can inhibit enzyme activities or bindings between PA, PB1 and PB2; 3.2 To evaluate in vitro anti-H1N1 effects of the selected compounds; 3.3 To detect in vitro cross antiviral effects of the selected compounds and the peptides against different subtypes of influenza virus; 3.4 To test potential toxicity of the selected compounds and the peptides in cell cultures; 3.5 To optimize the compounds by further screening analogues of the effective compounds; 3.6 To assess potential synergistic antiviral effects between effective compounds targeting different subunits of viral polymerase and the peptides; 3.7 To evaluate prophylactic and therapeutic anti-influenza effects of the selected compounds in established animal models and the peptides; 3.8 To study antiviral mechanism of the selected compounds and the peptides.
