Objectives
1 CPL for selective desulfurization. We aim to develop a strategy for selective desulfurization for chemical protein synthesis 2 CPL with thiolated amino acid and selenocysteine. Usage of the thiolated amino acids and selenocysteine will be explored for CPL. 3 CPL-solubilizing tags. We aim to introduce solubilizing tags via the salicylaldehyde ester moiety to realize CPL-solubilizing modification 4 Long-range thiol participation pathway analogous to CPL. Based on the CPL working principle, we will explore several strategies, including a relayed Lys acylation, an auxiliary-based ligation and intermolecular thiol-assisted peptide cyclization.
