Objectives
Osteoporosis is a prevalent disease characterized by low bone mass and deterioration in microarchitecture and bone strength with a consequent increased risk of fragility fracture. The aetiology of osteoporosis is multifactorial and complex. It has been established that genetic factors play an important role in disease determination. Using a genome-wide association study (GWAS) approach, more than 66 well validated loci have been found to be associated with variation in bone mineral density (BMD). Nonetheless whether rare variants (with frequency<1%), especially nonsynonymous variants, contribute this variation is largely unknown.
