Objectives
1 Develop a new method to prepare peptide salicylaldehyde esters via Boc-SPPS. 2 Develop on-resin Ser/Thr ligation and traceless on-resin ligation-mediated peptide cyclization. 3 Develop a homoserine ligation enabling the formation of Xaa-Asp at the ligation site. 4 Develop a method to improve Ser/Thr ligation using a solubilizing salicylaldehyde ester. 5 Develop new methods and strategies facilitating successive Ser/Thr ligation and apply this to the synthesis of the centra protein. For N to C Ser/Thr ligation, we will introduce a peptide salicylaldehyde
