AbstractOrexin is a hypothalamic neuropeptide associated with motor function of the vestibular nucleus (VN). In adult rats, deficiency of orexin was reported to decrease muscle strength and deteriorate vestibular-mediated behaviors. In early developing rats, we demonstrated that acute treatment of VN slices with orexin or its receptor agonist reduced GABA(A) receptor-mediated inhibitory postsynaptic currents. Also, shRNA knockdown of orexin in the lateral hypothalamus of neonatal rats enhanced acquisition of vestibular behaviors. Based on these findings, we further revealed the effects of orexin on developmental emergence of vestibular behaviors by perturbing orexin receptors in the VN of P1 rats. Behavioral performance was impaired in rats pretreated with orexin or its receptor agonists, but was enhanced in those pretreated with antagonists. Perturbation after P14 did not affect the behaviors, indicating a critical period for orexin in tuning the
